Yeast Elicitor Time Course
- A 5mg/ml yeast elicitor stock solution (in water) was used at a 1:100 dilution (2.5ml) for a final concentration of 50ug glucose equivalents/ml (Yeast elicitor prepared as described in Schumacher et al. 1987).
- 2.5ml of water was added to the control flasks
- The cultures were 11P, or in the 11th passage of subculturing
- The time course was initiated on Wednesday, May 8, 2002, with the final 48 hour culture harvested on Friday, May 10, 2002
- The 0 time point was initiated at 9:10AM; the final yeast elicitor treatment was done by 9:26AM (16 minutes from treatment start to treatment finish)
- The methyljasmonate internal controls were initiated at 9:29, 9:30, and 9:30AM
- The UV internal controls were initiated at 11:19 and 11:27AM
- All time point harvests were relatively uneventful and as scheduled/planned EXCEPT the 48 hour time point in which the lab lost house vacuum. This problem was overcome by manually applied vacuum; the samples were a bit wetter than the other time points
